目的:测定布洛芬糖浆剂的药动学及相对生物利用度。方法:8名健康男性志愿者单剂量随机交叉po400mg布洛芬后,采用HPLC测定血浆药物浓度。结果:口服糖浆剂及片剂后血药浓度达峰时间分别为(0.86±0.30)h和(1.78±0.34)h,峰浓度分别为(48.9±9.71)μg·ml-1和(44.2±8.23)μg·ml-1,药时曲线下面积分别为(175.1±31.9)μg·h·ml-1和(178.6±23.2)μg·h·ml-1。结论:统计分析表明,糖浆剂较片剂达峰时间短,吸收快。布洛芬糖浆剂相对生物利用度为(98.3±14.5)%。
Objective: To determine the pharmacokinetics and relative bioavailability of ibuprofen syrup. Methods: Eight healthy male volunteers were randomized to a single dose of po400mg ibuprofen for determination of plasma drug concentration by HPLC. Results: The peak plasma concentrations of oral syrup and tablets were (0.86 ± 0.30) h and (1.78 ± 0.34) h, respectively, with peak concentrations of (48.9 ± 9) 71.1 μg · ml -1 and (44.2 ± 8.23) μg · ml -1, respectively, and the areas under the drug-time curve were (175.1 ± 31.9) μg · h · ml -1 and (178 .6 ± 23.2) μg · h · ml-1. Conclusion: Statistical analysis shows that the syrup has a faster absorption than the peak time of the tablet. The relative bioavailability of ibuprofen syrup was (98.3 ± 14.5)%.
