目的研究VEGF siRNA对中国仓鼠肺细胞(CHL)的染色体畸变作用。方法采用25、50和100nmol/L的VEGF siRNA转染中国仓鼠的肺细胞,分别观察24h、48h后的染色体畸变情况。结果在VEGF siRNA转染24小时,仅100nmol/L VEGF siRNA产生可疑阳性反应,其染色体的畸变率为6%;在VEGF siRNA转染48小时后,50nmol/L的VEGF siRNA和100nmol/L的VEGF siRNA均产生可疑阳性反应,染色体的畸变率分别为6%和10%。而25nmol/L的VEGF siRNA无论是在转染后24h还是48h,均未产生染色体的畸变作用。VEGF siRNA产生的染色体畸变类型有断裂、双着丝粒、多倍体、裂隙、三射体。结论 100nmol/L的VEGF siRNA分子可引起CHL细胞产生染色体畸变。
Objective To investigate the role of VEGF siRNA in chromosomal aberration of Chinese hamster lung cells (CHL). Methods The lung cells of Chinese hamster were transfected with VEGF siRNA at 25, 50 and 100 nmol / L, and the chromosomal aberrations were observed 24h and 48h respectively. Results Only 100 nmol / L VEGF siRNA produced a suspicious positive response at 6 hours after transfection with VEGF siRNA. The chromosome aberration rate was 6%. After 48 hours of VEGF siRNA transfection, 50 nmol / L VEGF siRNA and 100 nmol / L VEGF The siRNAs all gave rise to a suspicious positive reaction with chromosome aberrations of 6% and 10%, respectively. However, 25nmol / L VEGF siRNA did not produce chromosome aberration either 24h or 48h after transfection. The types of chromosomal aberrations produced by VEGF siRNA are broken, dicentric, polyploidy, fissure, and the three emitters. Conclusion 100nmol / L VEGF siRNA molecules can cause chromosomal aberrations in CHL cells.
