用分子生物学的实验方法对非胰岛素依赖型糖尿病(NIDDM)患者的胰岛素基因进行检测。从NIDDM患者外周血的白细胞中提取DNA,以胰岛素基因5′-末端上游的某一DNA序列为引物,对特定的靶序列进行体外扩增,即TaqDNA聚合酶链式反应(PCR),将其扩增产物进行电泳分离,在紫外分析仪下观察结果并与正常人对照,发现16例NIDDM患者的胰岛素基因有所改变,提示胰岛素基因改变可能是导致NIDDM的原因之一。
Insulin gene in patients with non-insulin dependent diabetes mellitus (NIDDM) was tested by molecular biology method. DNA from peripheral blood leukocytes of patients with NIDDM was extracted, and a certain DNA sequence upstream of the 5’-end of the insulin gene was used as a primer to amplify a specific target sequence in vitro, that is, Taq DNA polymerase chain reaction (PCR) The amplified products were separated by electrophoresis and observed under ultraviolet spectrophotometer. Compared with normal controls, 16 cases of NIDDM patients had insulin gene changed, suggesting that insulin gene alteration may be one of the reasons leading to NIDDM.
