目的 研究不同温度热诱导后 He La 细胞热休克蛋白(heat shock protein70, H S P70)蛋白动态表达。方法 He La 细胞经过不同温热(41℃ 1 h、42℃ 1 h 、43℃ 1 h、45℃ 30 m in)的诱导,以 S D S P A G E及 Western 印迹方法检测细胞裂解液中 H S P70 蛋白表达。结果 热诱导后 H S P70 蛋白表达的高峰出现时间随诱导温度的提高逐渐后移(如:41℃ 8 h、43℃ 16 h),且 41℃、42℃热诱导后蛋白表达于诱导后 8~16 h 期间有持续高水平表达,而45℃ 30 m in 热诱导细胞大部分于诱导后即时死亡,且仅有低水平的 H S P70 表达;高表达后 H S P70 一般于诱导后24~32 h 始恢复至正常细胞表达水平。结论 温热诱导(41℃ 1 h)短时间内即可产生高表达量 H S P70,而较激烈热诱导(43℃ 1 h、45℃ 30 m in)仅产生少量延迟表达的 H S P70 蛋白,且导致大量 He La 细胞死亡,此为临床肿瘤温热治疗提供了理论依据。
Objective To investigate the dynamic expression of heat shock protein70 (HSP70) in He La cells after heat induction at different temperatures. Methods HeLa cells were induced by different warming (41°C for 1 h, 42°C for 1 h, 43°C for 1 h, and 45°C for 30 min). Cell lysates were detected by S D S P A G E and Western blotting methods.
