目的研究新疆哈萨克族食管癌中Cdc42基因启动子区甲基化状态与其mRNA表达改变的关系。方法用RT-PCR方法测定mRNA表达,甲基化特异PCR方法检测DNA启动子区甲基化状态。结果在20例癌组织中,有16例表达阳性,占80%,20例癌旁组织中有13例表达,占65%,其中癌组织表达量明显高于癌旁组织的有10例,占50%,癌旁组织表达量高于癌组织的有5例,占25%。肿瘤组织和正常组织该基因启动子区见甲基化条带,未见非甲基化条带。结论Cdc42基因在肿瘤组织和正常组织均表现为高甲基化状态,新疆哈萨克族食管癌中Cdc42基因mRNA表达增强可能与该基因的甲基化状态没有直接关系,另存在其他机制,有待进一步研究。
Objective To study the relationship between methylation status of Cdc42 promoter and its mRNA expression in Kazakh esophageal cancer in Xinjiang. Methods The expression of mRNA was detected by RT-PCR and the methylation status of DNA promoter was detected by methylation-specific PCR. Results Of the 20 cases of cancerous tissues, 16 cases were positive, accounting for 80%. Thirteen of 20 cases of paracancerous tissues were expressed, accounting for 65%, of which 10 cases were significantly higher than adjacent tissues 50%, the expression of paracancerous tissue is higher than that of cancer tissue in 5 cases, accounting for 25%. The tumor tissue and normal tissue promoter region of the gene to see methylated bands, no non-methylated bands. Conclusions The Cdc42 gene is highly methylated in both tumor and normal tissues. The increased expression of Cdc42 mRNA in Xinjiang Kazakh esophageal carcinoma may not be directly related to the methylation status of the gene. There are other mechanisms that need further study.
