目的研究大孔吸附树脂和超声波辅助解吸法纯化大田基黄总黄酮的工艺条件。方法比较11种大孔吸附树脂对大田基黄总黄酮的吸附与解吸附特性,筛选出性能最好的树脂进行后续研究,结合超声波考察影响解吸率的因素:超声波解吸时间、乙醇的体积分数及其用量。结果 D101型大孔吸附树脂在超声波的辅助解吸下,对大田基黄中总黄酮具有较好分离能力,最佳纯化工艺条件为:超声处理时间为30 min,乙醇的体积分数为60%,固液比1 g∶7 m L。结论采用本法分离大田基黄中的总黄酮成分简单可行。
Objective To study the conditions of purifying total flavonoids of Radix Oroxidene by macroporous adsorption resin and ultrasonic assisted desorption. Methods The adsorption and desorption characteristics of 11 macroporous adsorption resins on total flavonoids of Rhizoma Picrorhiza were compared. The best resin was screened out for follow-up study. The effects of ultrasonic desorption time, the volume fraction of ethanol, Its dosage. Results Under ultrasonic assisted desorption, D101 macroporous resin had good separation ability for total flavonoids in Radix et Rhizoma Rhei. The optimal purification conditions were as follows: the time of ultrasonic treatment was 30 min, the volume fraction of ethanol was 60% Liquid ratio 1 g: 7 m L. Conclusion This method is simple and feasible for the separation of flavonoids from Radix et Rhizoma.
