目的:研究定心方对缺血再灌注大鼠心肌MAPK通路的调节作用。方法:将40只雄性Wistar大鼠随机分为假手术组、模型组、定心方高、中、低剂量组,应用结扎(15min)和松开(30min)左冠状动脉前降支的方法制作缺血再灌注模型。应用免疫印迹技术检测MAPK通路中磷酸化ERK1/2、JNK1/2、P38蛋白的表达,并记录再灌注过程中Ⅱ导联心电图。结果:与模型组相比,其余各组的pJNK1/2和pP38表达及心律失常评分显著降低(P<0.05);定心方用药组的pERK1/2蛋白表达显著高于假手术组和模型组(P<0.001),而两组间无显著差异(P>0.05)。结论:定心方可以通过调节MAPK通路的激活,减轻大鼠心肌缺血再灌注损伤。
Objective: To study the regulation effect of Dingxin Recipe on MAPK pathway of myocardial ischemia-reperfusion in rats. Methods: Forty male Wistar rats were randomly divided into sham-operated group, model group, high-dose and low-dose group of middle-dose and low-dose group. They were made by ligation (15 min) Ischemia-reperfusion model. The expression of phosphorylated ERK1 / 2, JNK1 / 2 and P38 proteins in MAPK pathway was detected by Western blotting, and the Ⅱ lead electrocardiogram was recorded during the reperfusion. Results: Compared with the model group, pJNK1 / 2 and pP38 expression and arrhythmia score were significantly decreased in the other groups (P <0.05). The expression of pERK1 / 2 in the Shangxin Recipe group was significantly higher than that in the sham operation group and model group (P <0.001), but no significant difference between the two groups (P> 0.05). Conclusion: Dingxin Recipe can attenuate myocardial ischemia-reperfusion injury in rats by regulating the activation of MAPK pathway.
