人造的氧运载体(人造红细胞)可分为能溶解氧的化合物氟化碳和天然血红蛋白溶液或其衍生物两类。利用血红蛋白的人造红细胞氧结合力高,可长期保存。因此作者等研制了氧运载力更高的吡啶醛化的血红蛋白溶液(PLP-Hb)。按Benesch等的方法由新鲜红细胞或过期红细胞制备血红蛋白游离基质(SFH)溶液,系将红细胞用生理盐水反复洗涤3次,用2倍量水溶血,加5%氯化钠制成等张液。再经2800G离心30分钟,取上清液用微孔滤膜(0.45μm)过滤,然后用葡萄糖(1g/L)和维生素C溶液反复透析,制得SFH溶液。
Artificial oxygen carriers (artificial red blood cells) can be divided into dissolved oxygen compounds, carbon fluoride and natural hemoglobin solution or two types of derivatives. Artificial erythrocytes that utilize hemoglobin have high oxygen binding capacity and can be stored for long periods of time. Therefore, the authors developed a more oxygen-loaded pyridaldehyde hemoglobin solution (PLP-Hb). The hemoglobin free matrix (SFH) solution was prepared from fresh erythrocytes or expired erythrocytes according to the method of Benesch et al. The erythrocytes were repeatedly washed three times with normal saline, and then equilibrated with 2% aqueous hemolysis and 5% sodium chloride. After centrifugation at 2800G for 30 minutes, the supernatant was filtered through a microporous membrane (0.45 μm) and then dialyzed repeatedly with glucose (1 g / L) and vitamin C solution to prepare SFH solution.
