目的:探讨中药复方参乌冠心颗粒剂防治冠心病心肌缺血的分子作用机制。方法采用异丙肾上腺素连续多点皮下注射,复制大鼠心肌缺血动物模型。运用DNA末端标记(TUNEL)法,观察复方参乌冠心颗粒剂对缺血损伤大鼠心肌细胞凋亡的影响,并运用免疫组化的方法观察其对细胞凋亡相关基因Fas、Bcl-2蛋白表达的影响。结果①参乌冠心颗粒剂有明显的抗心肌缺血作用。②参乌冠心颗粒剂可显著下调Fas基因的蛋白表达,上调Bcl-2基因的蛋白表达,明显抑制和阻断心肌细胞发生凋亡。③参乌冠心颗粒剂可能通过抑制和阻断心肌细胞过度凋亡的发生而起到保护心肌的作用。结论通过调节缺血心肌Fas、Bcl-2基因的蛋白表达从而抑制和阻断细胞凋亡的发生,说明中药复方参乌冠心颗粒剂具有防治冠心病心肌缺血损伤的分子作用机制。
Objective: To explore the molecular mechanism of Shenfu Guanxin granules in prevention and treatment of myocardial ischemia in coronary heart disease. Methods Rat models of myocardial ischemia were established by continuous subcutaneous injection of isoproterenol. The effect of compound Shenwu Guanxin granule on cardiomyocyte apoptosis induced by ischemic injury was observed by TUNEL method. The apoptosis-related genes Fas and Bcl-2 were observed by immunohistochemistry. The effect of protein expression. Results Shenshen Guanxin granules had obvious anti-ischemic effect. Shenshen Guanxin granule can significantly down-regulate Fas gene protein expression, up-regulate Bcl-2 gene protein expression, and obviously inhibit and block cardiac myocyte apoptosis. Shenshen Guanxin granules may play a role in myocardial protection by inhibiting and blocking the occurrence of excessive apoptosis of myocardial cells. Conclusion By regulating the protein expression of Fas and Bcl-2 gene in ischemic myocardium, the occurrence of apoptosis is inhibited and blocked. It is concluded that Shenwu Guanxin granules have the molecular mechanism of prevention and treatment of ischemic injury of coronary heart disease.
