目的:验证益髓通经方经醇提后的药渣抗实验性自身免疫性神经炎(EAN)的活性,并优化其提取工艺。方法:建立大鼠EAN模型,将造模成功的大鼠随机分成模型组、免疫球蛋白组和给药组,按相应方法给药,用电子显微镜观察各组大鼠周围神经病理形态学超微结构;以干膏质量和蛋白提取率的综合评分为指标,采用正交试验法,选取加水量、提取时间和提取次数为考察因素,优化益髓通经方中水溶性成分的提取工艺。结果:给药组大鼠髓鞘脱失的情况得到明显改善,有髓纤维髓鞘板层结构整齐,胶原纤维聚集及有髓神经纤维新生等改变;最佳提取工艺为加8倍量水提取3次,每次3 h。结论:益髓通经方水溶性成分具有治疗EAN的效果;优选的提取工艺操作简单、稳定性好,适合于产业化生产。
OBJECTIVE: To verify the activity of Yisui Tongjing Decoction anti-experimental autoimmune neuritis (EAN) after alcohol extraction and to optimize its extraction process. Methods: EAN model was established in rats. The successful rats were randomly divided into model group, immunoglobulin group and administration group. The rats were administered with corresponding methods. The ultrastructure of peripheral nerve pathomorphology Structure; taking the comprehensive score of dry mass and protein extraction rate as the index, using orthogonal test method, selecting the amount of water added, the extraction time and the number of extraction as investigation factors, and optimizing the extraction process of water soluble components in Yisui Tongjing prescription. Results: The demyelination of the rats in the treatment group was significantly improved. The structure of the myelinated myelin sheath was well-proportioned, the aggregation of collagen fibers and the regeneration of myelinated nerve fibers were the best. The optimal extraction process was 8-fold water extraction 3 times, 3 h each. Conclusion: The water-soluble components of Yisutongjing Recipe have the effect of treating EAN. The optimal extraction process is simple, stable and suitable for industrial production.
