目的:探讨四物汤有效成分与脂质体模拟生物膜的相互作用,以了解其有效成分体内吸收的情况。方法:采用平衡透析与高效液相色谱-质谱联用技术,对四物汤有效成分与脂质体模拟生物膜相互作用进行分析;同时考察了透析时间、脂质体溶液的浓度和缓冲溶液p H对四物汤有效成分与模拟生物膜相互作用的影响。结果:四物汤中有9个成分与脂质体相互作用明显,通过对照品对照及LC-MS方法鉴定了其中6个成分,分别是香草酸、芍药内酯苷、毛蕊花糖苷、芍药苷、洋川芎内酯Ⅰ、肉桂酸;模拟生物膜的浓度对四物汤有效成分与模拟生物膜相互作用影响较大,随着浓度增加,色谱峰面积明显减小,符合被动吸收规律;p H对其中4个成分有明显影响。结论:脂质体平衡透析液质联用色谱法可作为快速筛选中药潜在的活性成分的有效方法,为药效物质基础的进一步研究提供参考。
OBJECTIVE: To investigate the interaction between active ingredients of Siwu Tang and liposome mimic biofilms to understand the absorption of active ingredients in vivo. Methods: The interaction between active ingredients of Siwu Decoction and mimic biofilm was analyzed by equilibrium dialysis and high performance liquid chromatography-mass spectrometry. Results: There were 9 components in Siwu Decoction, which had obvious interaction with liposome. Six components were identified by LC-MS and control samples, which were vanillic acid, paeoniflorin, abacus glycoside, paeoniflorin, The concentration of simulated biofilm had a significant effect on the interaction between the active ingredients of Simwu Decoction and the simulated biofilm, with the increase of the concentration, the peak area of the chromatogram obviously decreased, which accorded with the passive absorption law; Four of these ingredients have a significant effect. Conclusion: Liposomal equilibrium dialysate chromatography-mass spectrometry can be used as an effective method for rapid screening of potential active ingredients of traditional Chinese medicine for reference for further study on the basis of pharmacodynamic substances.
