目的:探讨健脾益气活血汤通过血管紧张素转换酶2(ACE2)、血管紧张素1-7(Ang 1-7)-MAS受体、蛋白激酶B(AKT)信号通路调节自发性高血压大鼠(SHR)血管活性因子释放的作用。方法:将60只24周龄SHR分为SHR组(给予蒸馏水)、培哚普利组(0.4 mg·kg-1·d-1)、培哚普利联合中药组(培哚普利0.4 mg·kg-1·d-1+健脾益气活血汤20 g·kg-1·d-1,中加西组)、健脾益气活血汤高、中、低剂量组(40,20,10 g·kg-1·d-1),每组10只,以同龄同种系正常血压的京都种大鼠(WKY)10只作为WKY组(给予蒸馏水)。无创血压计监测大鼠尾动脉收缩压;治疗6周后腹主动脉取血测定血浆Ang(1-7)含量,取肾脏提取总RNA及总蛋白,RT-PCR法检测ACE2,MAS mRNA含量表达,Western blot法检测AKT蛋白表达水平。结果:SHR组ACE2,Ang(1-7),MAS,AKT含量与WKY组相比明显减少(P<0.01);培哚普利组、中加西组收缩压与SHR组相比明显降低(P<0.01);培哚普利组、中加西组、中药高剂量组ACE2,Ang(1-7),MAS,AKT含量明显高于SHR组(P<0.01)。结论:健脾益气活血汤通过ACE2-Ang(l-7)-MAS-AKT信号通路平衡血管内皮舒缩因子的分泌与释放,发挥改善血管内皮功能及抗高血压作用。
Objective: To investigate the effects of Jianpi Yiqi Huoxue Decoction on regulating spontaneous hypertension through angiotensin converting enzyme 2 (ACE2), angiotensin 1-7 (MAP-1-7) -MAS receptor and AKT signal pathway Role of Vasoactive Factor Release in Rat (SHR). METHODS: Sixty SHR (24 weeks old) SHR group (distilled water), perindopril group (0.4 mg · kg -1 · d -1), perindopril combined with traditional Chinese medicine group (perindopril 0.4 mg Kg-1 · d-1 + Jianpi Yiqi Huoxue Decoction 20g · kg-1 · d-1, Zhongjiaxi group), and Jianpi Yiqi Huoxue Decoction high, medium and low dose groups g · kg-1 · d-1). Ten rabbits in each group were enrolled, and 10 WKY rats of the same age with the same type of normal blood pressure were used as WKY group (given distilled water). Non-invasive sphygmomanometer was used to monitor systolic pressure of caudal artery in rats. Blood was taken from abdominal aorta for 6 weeks to measure plasma Ang (1-7) content, total RNA was extracted from kidney and total protein was detected by RT-PCR. The protein expression of AKT was detected by Western blot. Results: The levels of ACE2, Ang (1-7), MAS and AKT in SHR group were significantly lower than those in WKY group (P <0.01). The systolic blood pressure in perindopril group and Zhongjiaxi group was significantly lower than that in SHR group <0.01). The contents of ACE2, Ang (1-7), MAS and AKT in perindopril group, Zhongjiaxi group and traditional Chinese medicine high dose group were significantly higher than those in SHR group (P <0.01). Conclusion: Jianpi Yiqi Huoxue Decoction can balance the secretion and release of vascular endothelium contraction and contraction factor through ACE2-Ang (1-7) -MAS-AKT signaling pathway and play an important role in improving vascular endothelial function and antihypertensive effect.
