目的 分离纯化鼠源性抗肥大细胞FcεRIα(高亲和力的IgE的Fc段受体)的单克隆抗体。方法 先培养ER-E5.3细胞,获得大量的含抗肥大细胞FcεRIα的单克隆抗体的培养上清。然后应用A蛋白-Sphrose CL-4B制备亲和层析柱,分离纯化细胞培养上清中的单克隆抗体。结果分离纯化的单克隆抗体纯度高,活性强。结论应用A蛋白可以很好分离纯化细胞培养上清中的鼠源性抗肥大细胞FcεRIα的单克隆抗体。
OBJECTIVE To isolate and purify monoclonal antibody against murine anti-mast cell FcεRIα (high affinity IgE Fc receptor). Methods ER-E5.3 cells were cultured and a large number of culture supernatant of monoclonal antibody containing anti-mast cell FcεRIα was obtained. Then, the protein A-Sphrose CL-4B was used to prepare the affinity chromatography column, and the monoclonal antibody in the cell culture supernatant was separated and purified. Results The monoclonal antibodies isolated and purified showed high purity and activity. Conclusion The monoclonal antibody against murine anti-Mast cell FcεRIα in cell culture supernatant can be well separated and purified using protein A.
