目的:探讨PD98059对慢性髓细胞白血病信号转导的作用及其作用机制。方法:将PD98059加入慢性髓细胞白血病细胞系K562细胞中,通过细胞活力、DNA合成、集落形成和MAPK活性的变化,观察PD98059对慢性髓细胞白血病信号转导的作用。结果:PD98059可明显抑制K562细胞活力、DNA合成、集落形成和MAPK活性(与对照组比较,P<0.05),其抑制作用具有浓度依赖性。结论:PD98059可通过阻断Ras-MAPK信号通路而发挥其抑制作用。Ras-MAPK通路极有可能成为CML治疗的新靶点。
Objective: To investigate the effect of PD98059 on signal transduction of chronic myeloid leukemia and its mechanism. Methods: PD98059 was added to K562 cells. The effects of PD98059 on signal transduction of chronic myelogenous leukemia were observed by cell viability, DNA synthesis, colony formation and MAPK activity. Results: PD98059 could significantly inhibit K562 cell viability, DNA synthesis, colony formation and MAPK activity (P <0.05 compared with the control group), and its inhibitory effect was concentration-dependent. Conclusion: PD98059 can exert its inhibitory effect by blocking the Ras-MAPK signaling pathway. Ras-MAPK pathway is likely to become a new target for the treatment of CML.
