The genetic polymorphisms of the D19S400 locus in Chinese Han, German, Slovak and Afro-American populations and the differences among the three major human races in China were analyzed by PCR. Of the four groups, 620 were surveyed, 11 alleles were found and 47 genotypes were observed. The observed heterozygosity of each group was 0.78 ~ 0.88, and the probability of personal identification was 0.93850 ~ 0.9664. The frequency distribution of genotypes of the four groups was in accordance with the Hardy-Weinberg equilibrium (P> 0.05). There was a significant difference in allele frequencies of the D19S400 locus between the three races (Mongolian, Caucasians, African Americans) P <0.01). The results showed that the D19S400 locus has high value in population genetics and forensic personal identification
