目的研究大蒜素联合阿糖胞苷对体外培养的人红白血病细胞株(K562)增殖及细胞凋亡的影响。方法处于对数生长期的K562细胞,给予阿糖胞苷、大蒜素及两药联合处理细胞,经瑞氏染色观察细胞形态学变化,并以MTT比色法检测细胞增殖活性及药物相互作用方式。结果单用大蒜素或阿糖胞苷均可抑制K562增殖并促进其凋亡,并呈时-效和量-效依赖性;联合组对K562增殖及凋亡的影响明显增强。大蒜素对Ara-c的增效倍数为2.22,且二者的作用方式为协同作用。结论大蒜素与阿糖胞苷联用具有明显的抗肿瘤作用,大蒜素可提高阿糖胞苷抗白血病的敏感性,从而降低临床上阿糖胞苷用药剂量和细胞毒性,增强疗效;此为白血病的临床治疗提供了新思路。
Objective To study the effects of allicin combined with cytarabine on the proliferation and apoptosis of human erythroleukemia cell line (K562) cultured in vitro. Methods K562 cells in logarithmic growth phase were treated with cytosine arabinoside, allicin and the two drugs in combination. Wright’s stain was used to observe the morphological changes of cells. MTT assay was used to detect the cell proliferation and drug interactions . Results Allicin or cytarabine alone inhibited the proliferation and promoted the apoptosis of K562 cells in a time-and dose-dependent manner. The combination group significantly enhanced the proliferation and apoptosis of K562 cells. The synergistic ratio of allicin to Ara-c is 2.22, and the mode of action of both is synergistic. Conclusion Allicin combined with cytarabine has obvious anti-tumor effect, allicin can increase the sensitivity of cytarabine anti-leukemia, thereby reducing the dose and cytotoxicity of cytarabine clinical and enhance the efficacy of this Clinical treatment of leukemia provides a new idea.
