有研究者用L-[1-13C]苯丙氨酸呼气试验(PheBT)发现肝病患者摄入L-[1-13C]苯丙氨酸(13C-phe)后13CO2排除能力下降。我们对乙型肝炎后肝硬化患者的相关研究也取得有意义的结果。由于13CO2呼气试验的内在机制以及影响试验结果的因素并不十分明确,从而限制了呼气试验在临床的广泛应用。我们采用自行设计的小动物呼气试验模型进行大鼠PheBT,以验证该实验方法的可行性和有效性,并提供有效的试验参数。一、材料与方法1.实验动物:清洁级(SPF)SD大鼠,雄性,体重220- 230 g,由复旦大学实验动物中心提供,大鼠饲养在SPF级动物室中。观察1周后,选取体重280~290 g的大鼠20只,随机分为两组,每组10只。
Some researchers found that the depletion of 13CO2 in liver disease patients after their ingestion of L- [1-13C] -phenylalanine (13C-phe) was found by the L- [1-13C] phenylalanine breath test (PheBT). Our research on patients with posthepatitic cirrhosis has also yielded meaningful results. Because of the inherent mechanism of 13CO2 breath test and the factors affecting the test results are not very clear, which limits the extensive clinical application of breath test. We designed a rat model of PheBT using a small animal breath test model designed to verify the feasibility and effectiveness of this experimental method and provide valid experimental parameters. I. Materials and Methods 1. Experimental animals: SPF SD rats, male, weighing 220-230 g, were provided by Experimental Animal Center of Fudan University and rats were housed in SPF animal chambers. One week after the observation, twenty rats weighing 280-290 g were randomly divided into two groups of 10 rats.
