目的:探讨决明子、黄芪对大鼠棕色脂肪线粒体内膜解偶联蛋白(UCP)活性与含量的影响。方法:用~3H 标记的鸟嘌呤核苷三磷酸(GTP)与 UCP 相结合,用液体闪烁仪测定其放射性活性,经 ScatchardPlot 分析得出两者结合的解离常数(Kd)和最大结合量(Bmax)。结果:决明子组 Bmax、Kd 值与对照组比较无显著性差异;黄芪组较对照组 Bmax 增加1.29倍,Kd 值无改变。结论:决明子对棕色脂肪线粒体内膜 UCP含量增加及活性无明显影响;黄芪能使棕色脂肪线粒体内膜 UCP 含量增加,但不能使其活性增加。
Objective: To investigate the effects of cassia seed and astragalus membranaceus on activity and content of uncoupling protein (UCP) in mitochondrial inner membrane of rat brown fat. METHODS: The ~3H-labeled guanosine triphosphate (GTP) was combined with UCP. The radioactivity of the product was measured with a liquid scintillation detector. The dissociation constant (Kd) and the maximum binding amount were determined by Scatchard Plot analysis. Bmax). Results: There was no significant difference in Bmax and Kd values between the cassia seed group and the control group. The Bmax of the astragalus group was 1.29 times higher than that of the control group, and there was no change in the Kd value. CONCLUSION: Cassia seed has no significant effect on the increase of UCP content and activity in mitochondrial inner membrane of brown fat; Astragalus membranaceus can increase the UCP content in inner membrane of brown fat mitochondria, but it can not increase its activity.
