目的:应用SRB法探讨不同药物对骨髓间充质干细胞(MSCs)、肝癌细胞(HC)和HepG2细胞的药敏差异.方法:雌性清洁级SD大鼠20只,应用二乙基亚硝胺饲喂法建立大鼠原发性肝癌模型,选取诱癌成功大鼠8只分别原代培养HC与MSCs,同时培养HepG2细胞系细胞,SRB法进行药物敏感实验,观察药物敏感性的差异.结果:对阿霉素、顺铂、洛拉曲克的敏感度从高到低依次为HepG2细胞,HC,MSCs.而丝裂霉素药敏显示,3种细胞药物敏感度从高到低依次为HepG2,MSCs,HC.结论:HC药敏实验与HepG2细胞和MSCs存在一定异同.
OBJECTIVE: To investigate the drug sensitivity of different drugs to bone marrow mesenchymal stem cells (MSCs), hepatoma cells (HC) and HepG2 cells by SRB method.Methods: Twenty female SD rats were divided into two groups: Methods The primary hepatocarcinoma model was established in rats, and 8 primary cultured rats were used to culture HC and MSCs respectively, HepG2 cells were also cultured and SRB method was used to detect drug sensitivity.Results: The sensitivity to doxorubicin, cisplatin and nolatrexed was HepG2, HC and MSCs from high to low, while the susceptibility to mitomycin showed that the sensitivity of three kinds of cells from high to low were HepG2 , MSCs, HC.Conclusion: HC susceptibility test has some similarities and differences with HepG2 cells and MSCs.
