目的:探讨He-Ne激光对TNF基因的调控作用。方法:用斑点杂交及Northern 杂交技术观测激光对肿瘤患者TNF基因表达的影响,采用24m W He-Ne激光照射荷瘤小鼠及肿瘤患者脾区(瘤区),照射前后分别取脾细胞及外周血淋巴细胞,从中提取RNA。通过双抗体夹心ELLSA 法及受体吸收试验,观测激光照射对带瘤机体TNF受体及TNF活性的影响。结果:带瘤机体TNFm RNA 及TNF活性明显降低,血清及培养上清中可溶性TNF受体(sTNFR)的表达量增高。但经激光照射后TNFm RNA 表达量显著增高,TNF活性增强,而sTNFR 含量减少,瘤块明显减小。照射前后结果比较,差异有高度显著性(P< 0.01)。结论:He-Ne激光照射具有促进TNFm RNA 表达及抗肿瘤的作用
Objective: To investigate the regulation of TNF gene by He-Ne laser. METHODS: Dot blot hybridization and Northern blot were used to observe the effect of laser on TNF gene expression in tumor patients. 24m W He-Ne laser was used to irradiate tumor-bearing mice and spleen (tumor area) of tumor patients. Splenocytes and peripheral were taken before and after irradiation. Blood lymphocytes, from which RNA is extracted. The effect of laser irradiation on the tumor TNF receptor and TNF activity was observed by double antibody sandwich ELLSA method and receptor absorption assay. RESULTS: The activity of TNFm RNA and TNF in the tumor-bearing mice was significantly decreased, and the expression of soluble TNF receptor (sTNFR) in the serum and culture supernatants was increased. However, after laser irradiation, the expression of TNFm RNA was significantly increased, TNF activity was increased, and sTNFR content was decreased, and tumor mass was significantly reduced. The difference between the results before and after irradiation was highly significant (P < 0.01). Conclusion: He-Ne laser irradiation has the effect of promoting TNFm RNA expression and anti-tumor effect
