目的 探讨人结直肠癌组织CDK4mRNA转录和蛋白表达水平及其临床意义。方法 通过Northernblot和Westernblot方法 ,分析 30例人结直肠癌和其相应正常粘膜组织中 ,CDK4基因mRNA转录和蛋白表达水平。结果 40 %的结直肠癌组织中CDK4mRNA转录水平增高 ,5 3 3%的癌组织蛋白表达水平增高。其表达情况与肿瘤分化程度有关 ,表达水平越高 ,分化程度越低 ,二者比较差异有显著性 (χ2 =13 0 5 ,P =0 0 2 3;χ2 =13 89,P =0 0 0 9)。结论 CDK4可能是结直肠癌的原癌基因 ,在肿瘤发生过程中起一定作用。可做为结肠癌基因治疗的理论依据。
Objective To investigate the expression and clinical significance of CDK4 mRNA transcription and protein expression in human colorectal cancer. Methods Northern blot and Western blot were used to analyze the transcription and protein expression of CDK4 mRNA in 30 human colorectal cancers and their corresponding normal mucosal tissues. Results The transcription level of CDK4 mRNA was increased in 40% of colorectal cancer tissues, and the expression of cancer tissue protein was increased in 533%. Its expression was related to the degree of tumor differentiation. The higher the expression level, the lower the degree of differentiation. There was a significant difference between the two groups (χ 2 =13 0 5, P =0 0 2 3; χ 2 =13 89, P =0 0 0 9). Conclusion CDK4 may be a proto-oncogene of colorectal cancer and play a role in tumorigenesis. It can serve as a theoretical basis for gene therapy for colon cancer.
