目的 探讨Bcl- 1基因重排 (即Bcl- 1/IgH基因重排 )在B细胞慢性淋巴细胞白血病中的发生情况及其临床意义。方法 我们对 13例慢性淋巴细胞白血病 (CLL)进行了Bcl- 1基因重排的检测 ;对照组为 10例骨髓正常者。运用半筑巢式多聚酶链反应 (PCR)技术对从外周血、骨髓中提取的基因组DNA进行扩增。结果 这 13例CLL均为B细胞型 ,在 13例CLL中检出 4例 (31% )的Bcl- 1基因重排 ;10例骨髓正常者的对照组中未检测出Bcl- 1/IgH基因重排。结论 (1)B细胞慢性淋巴细胞白血病常发生Bcl- 1基因重排 ,骨髓正常者未检出Bcl- 1/IgH基因重排。 (2 )发生Bcl- 1基因重排的B细胞慢性淋巴细胞白血病外周血中原始或幼稚淋巴细胞增多。
Objective To investigate the occurrence and clinical significance of Bcl-1 gene rearrangement (ie Bcl-1/IgH rearrangement) in B-cell chronic lymphocytic leukemia. Methods We performed Bcl-1 gene rearrangement in 13 patients with chronic lymphocytic leukemia (CLL). The control group included 10 normal bone marrow patients. Genomic DNA extracted from peripheral blood and bone marrow was amplified using a semi-nested polymerase chain reaction (PCR) technique. RESULTS: All 13 CLLs were of B-cell type. Four cases (31%) of Bcl-1 gene rearrangements were detected in 13 cases of CLL; Bcl-1/IgH genes were not detected in the control group of 10 cases of normal bone marrow. rearrange. Conclusions (1) The Bcl-1 gene rearrangement often occurs in B-cell chronic lymphocytic leukemia, and Bcl-1/IgH gene rearrangement is not detected in normal bone marrow. (2) Presence or premature lymphocytosis of peripheral blood of B-cell chronic lymphocytic leukemia with Bcl-1 gene rearrangement.
