本文报告了血浆血栓素B_2的放射免疫分析法。样品经酸化后用乙酸乙酯提取,再经石油醚提取,排除了脂肪干扰,提高了方法的准确性。抗血清以TXB_2-BSA为免疫原注射兔制备。[~3H]-TXB_2的比放射性为140ci/mmol。放射免疫分析采用非平衡法,提高了灵敏度。本法主要技术指标如下:标准曲线范围为2.5～160pg/管,三种不同浓度血浆批内CV分别为6.4、7.8、7.3％。提取示踪回收为90.3％,冷回收为92.3％。健康成人血浆测定值为127±48pg/ml。
This article reports the radioimmunoassay of plasma thromboxane B 2. Samples were acidified with ethyl acetate extraction, and then extracted with petroleum ether, ruled out fat interference and improve the accuracy of the method. Antiserum was prepared by injecting rabbit with TXB_2-BSA as an immunogen. The specific activity of [~ 3H] -TXB_2 was 140 ci / mmol. Radioimmunoassay using non-equilibrium method to improve the sensitivity. The main technical indicators are as follows: The standard curve range of 2.5 ~ 160pg / tube, three different concentrations of plasma within the CV were 6.4,7.8,7.3%. Tracer recovery was 90.3% for extraction and 92.3% for cold recovery. Plasma values for healthy adults were 127 ± 48 pg / ml.
