目的:制备抗鸡生肌蛋白单克隆抗体并鉴定其抗原识别特异性。方法:以重组的谷胱甘肽转硫酶-生肌蛋白融合蛋白为抗原,采用杂交瘤技术制备抗生肌蛋白单抗,并结合蛋白印迹及胶阻滞分析进行特异性鉴定。结果:所得6株杂交瘤中有一株分泌单抗(S45)只专一识别生肌蛋白,并可与DNA-生肌蛋白复合物结合,形成DNA-生肌蛋白-抗体超级复合物。结论:该单抗可用于胚胎发育过程生肌蛋白的表达及骨骼肌特异基因表达调控研究。
Objective: To prepare anti-chicken myosin monoclonal antibody and identify its antigen recognition specificity. Methods: Recombinant glutathione S-transferase-myogen fusion protein was used as antigen to prepare anti-live muscle myosin monoclonal antibody by hybridoma technique. Western blot and gel retardation analysis were used to identify the specific anti-myosin MAbs. Results: One monoclonal antibody (S45) secreted by one of the 6 hybridomas was identified specifically as myogenic protein and binds to the DNA-myosin complex to form a DNA-myogenin-antibody supercomplex. Conclusion: The McAb can be used to study the expression of myogenic protein and regulation of skeletal muscle-specific genes during embryonic development.
