目的:观察丹参多酚酸盐预处理对大鼠心肌缺血再灌注损伤的防护作用。方法:将健康成年雄性SD大鼠40只随机分为假手术组(S组)、缺血再灌注组(I/R组)、低剂量组(LD组)和高剂量组(HD组)。采用结扎大鼠左冠状动脉前降支(LAD)30min、恢复灌注120min制备大鼠心肌缺血再灌注模型。分别于药物输注前(T1),LAD阻断后30min(T2),LAD开放后30min(T3)、3h(T4)、6h(T5)、24h(T6)共6个时点测定血清一氧化氮合酶(NOS)、超氧化物岐化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GPX)、肌酸磷酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)及肌钙蛋白(cTnI)水平。结果:与I/R组比较,LD组和HD组血清CK-MB、LDH、cTnI、MDA及NOS水平显著降低,SOD与GPX含量显著上升(均P<0.05)。结论:丹参多酚酸盐预处理对大鼠心肌缺血再灌注损伤具有较好的防护作用。
Objective: To observe the protective effect of Salvia miltiorrhiza bunge polyphenols preconditioning on myocardial ischemia-reperfusion injury in rats. Methods: Forty healthy male Sprague Dawley rats were randomly divided into sham operation group (S group), ischemia reperfusion group (I / R group), low dose group (LD group) and high dose group (HD group). The model of myocardial ischemia-reperfusion in rats was established by ligation of left anterior descending coronary artery (LAD) in rats for 30 min and reperfusion for 120 min. Serum levels of nitric oxide were measured at 6 time points before drug infusion (T1), 30min (T2) after LAD blockade, 30min (T3), 3h (T4), 6h (T5) and 24h (T6) NOS, SOD, MDA, GPX, CK-MB, lactate Dehydrogenase (LDH) and troponin (cTnI) levels. Results: Compared with I / R group, the levels of serum CK-MB, LDH, cTnI, MDA and NOS in LD group and HD group were significantly decreased, and the contents of SOD and GPX were significantly increased (all P <0.05). Conclusion: Salviae miltiorrhizae polyphenols preconditioning has a good protective effect on myocardial ischemia-reperfusion injury in rats.
