目的:研究黑果小檗总花色苷(BHSTA)对小鼠小胶质细胞(BV2细胞)分泌炎性因子以及AD小鼠学习记忆能力的影响。方法:用Aβ_(25-35)诱导BV2细胞构建神经炎性反应模型,采用ELISA法检测BHSTA对BV2细胞分泌IL-1β、IL-6、TNF-α水平的影响;通过对小鼠侧脑室注射Aβ_(25-35)构建AD小鼠模型,采用Morris水迷宫试验评价BHSTA对AD小鼠学习记忆能力的影响。结果:Aβ_(25-35)诱导后,细胞上清液中IL-1β、IL-6、TNF-α明显升高(P<0.01),给予BHSTA后,细胞上清液中IL-1β、IL-6、TNF-α明显下降(P<0.05,P<0.01)。Morris水迷宫实验显示,BHSTA能够缩短小鼠水迷宫平台训练实验的潜伏期。结论:BHSTA对BV2细胞神经炎症模型具有明显保护作用,能够提高Aβ_(25-35)致痴呆小鼠的学习记忆能力。
Objective: To investigate the effect of total flavonoids of barberry Berberis (BHSTA) on inflammatory factors secreted by mouse microglia (BV2 cells) and the learning and memory abilities of AD mice. Methods: The model of neuroinflammation was established by inducing BV2 cells with Aβ_ (25-35). The effect of BHSTA on the levels of IL-1β, IL-6 and TNF-α secreted by BV2 cells was detected by ELISA. Aβ_ (25-35) was used to construct AD mouse model. Morris water maze test was used to evaluate the effect of BHSTA on learning and memory in AD mice. Results: The levels of IL-1β, IL-6 and TNF-α in the cell supernatant were significantly increased after Aβ_ (25-35) induction (P <0.01) -6, TNF-αdecreased significantly (P <0.05, P <0.01). Morris water maze test showed that BHSTA can shorten the incubation period of water maze test in mice. CONCLUSION: BHSTA has a significant protective effect on the BV2 cell model of neuroinflammation and can improve the learning and memory abilities of mice with dementia induced by Aβ_ (25-35).
