目的探讨生脉注射液对青春期大鼠睾丸扭转复位后Bcl-2/Bax蛋白表达的影响。方法 6周龄健康SD大鼠,♂,16只,建立单侧睾丸扭转复位模型。随机分为睾丸扭转复位生理盐水(对照)组和睾丸扭转复位生脉注射液(实验)组,每组8只。术后9周取双侧睾丸标本,HE染色后观察睾丸组织形态改变;免疫组织化学ABC法检测Bcl-2/Bax蛋白在睾丸组织中的表达。结果实验组生精小管管壁完整,生精上皮排列规则,细胞层次清晰;对照组生精小管管壁不完整,生精上皮变薄,细胞排列紊乱,层次减少,部分细胞可见核固缩,核碎裂崩解,可见多核细胞。与对照组相比,实验组睾丸组织Bcl-2蛋白表达增加(P<0.05),Bax蛋白表达减少(P<0.05)。结论生脉注射液可能通过增加Bcl-2蛋白和降低Bax蛋白的表达减轻细胞凋亡,对睾丸扭转复位后的损伤有保护作用。
Objective To investigate the effect of Shengmai Injection on the expression of Bcl-2 / Bax protein after adolescent testicular torsion in rats. Methods Healthy 6-week-old SD rats, ♂ and 16, were established to establish unilateral testicular torsion-reduction model. Randomly divided into testicular torsion reduction saline (control group) and testicular torsion reduction Shengmai injection (experimental group), 8 in each group. Bilateral testis specimens were obtained 9 weeks after operation and the morphology of testis was observed by HE staining. The expression of Bcl-2 / Bax protein in testis was detected by immunohistochemical ABC method. Results The seminiferous tubules of the experimental group had intact wall with regular arrangement of the seminiferous epithelium and clear cell layers. In the control group, the walls of the seminiferous tubules were incomplete, the seminiferous epithelium became thinner, the cells were disordered and the layers were reduced. Some cells showed nuclear pyknosis, Nuclear fragmentation disintegrates, showing multicellular cells. Compared with the control group, the expression of Bcl-2 protein in experimental group increased (P <0.05) and the expression of Bax decreased (P <0.05). Conclusion Shengmai injection may reduce apoptosis by increasing Bcl-2 protein and Bax protein expression, and have a protective effect on the damage after testicular torsion and reduction.
