目的观察有氧运动对肥胖大鼠脂肪组织锌α糖蛋白(ZAG)基因表达的影响及其与胰岛素敏感指数(ISI)的相关性,为肥胖及相关疾病的防治提供新思路。方法对饮食诱导的肥胖大鼠进行6周有氧运动干预,干预后检测大鼠体重增加值、内脏脂肪垫相对重量、空腹血糖(FBG)、血清甘油三酯(TG)、空腹血清胰岛素(FINS)含量及ISI,采用RT-PCR检测脂肪组织中ZAGmRNA表达水平。结果 (1)运动干预后,肥胖运动组(DIO-E)的体重增加值显著低于对照组(P0.01),与肥胖安静组(DIO-NE)相比虽无显著性差异,但呈下降趋势(P0.061)。(2)DIO-E组大鼠内脏脂肪垫相对重量、FBG,、FINS、TG含量显著低于DIO-NE组;而ZAGmRNA表达水平、ISI则高于对照组和DIO-NE组(P0.05,P0.01)。(3)大鼠脂肪组织ZAGmRNA表达水平和体重增加值、内脏脂肪垫相对重量、FBG、FINS呈显著性负相关(P0.05),与ISI呈高度正相关(r0.712,P0.01),和TG无显著相关(P0.05)。结论运动可以上调肥胖大鼠脂肪组织ZAGmRNA表达;ZAGmRNA表达增加可促进机体脂质代谢,提高胰岛素敏感性。
Objective To observe the effect of aerobic exercise on the gene expression of zinc α-glycoprotein (ZAG) in adipose tissue of obese rats and its correlation with insulin sensitivity index (ISI), and to provide a new idea for the prevention and treatment of obesity and related diseases. Methods Six-week aerobic exercise intervention was performed on diet-induced obese rats. After intervention, body weight gain, visceral fat pad weight, fasting blood glucose (FBG), serum triglyceride (TG), fasting serum insulin ) Content and ISI, the expression of ZAG mRNA in adipose tissue was detected by RT-PCR. Results Compared with control group (P <0.01), weight gain of obese exercise group (DIO-E) was significantly lower than that of obese quiet group (DIO-NE) Decline trend (P0.061). (2) The relative weight of visceral fat pad, FBG, FINS and TG contents in DIO-E group were significantly lower than those in DIO-NE group, while the expression of ZAG mRNA and ISI in control group and DIO-NE group were significantly higher than those in DIO-NE group , P <0.01). (3) There was a significant negative correlation between ZAG mRNA expression and body weight gain, visceral fat pad weight, FBG and FINS (P <0.05), and was positively correlated with ISI (r0.712, P <0.01) , No significant correlation with TG (P> 0.05). Conclusion Exercise can increase the expression of ZAG mRNA in adipose tissue of obese rats. The increased expression of ZAG mRNA can promote lipid metabolism and increase insulin sensitivity.
