目的:本实验旨在探讨一次性力竭运动后不同时相,大鼠海马神经元发生凋亡的情况及凋亡调控基因对其的调控作用。方法:雄性SD大鼠28只,随机分为对照组,运动后即刻组,运动后12小时组和运动后24小时组。运动组在力竭跑台训练后即刻、12h、24h分别取大鼠大脑的海马组织,用免疫组织化学法检测Bcl-2、Bax蛋白的表达,Image Pro-Plus分析Bcl-2、Bax的积分光密度值。结果:力竭运动可引起海马神经元中Bcl-2蛋白表达降低而Bax蛋白表达升高,当两者的比值发生失衡出现显著下降,即可促进海马神经元凋亡。说明海马神经元中凋亡调控基因Bcl-2、Bax在力竭运动后不同时相对细胞凋亡的发生有明显的调控作用。
OBJECTIVE: To investigate the apoptosis of hippocampal neurons and the regulatory role of apoptosis-regulating genes in rats at different phases after one-time exhaustive exercise. Methods: Totally 28 male SD rats were randomly divided into control group, immediate after exercise group, 12 hours after exercise group and 24 hours after exercise group. The hippocampal tissue of rat’s brain was taken at 12h and 24h after exercise, and the expression of Bcl-2 and Bax protein was detected by immunohistochemical method. The integral of Bcl-2 and Bax was analyzed by Image Pro-Plus Optical density value. Results: Exhausted exercise could decrease the expression of Bcl-2 protein and Bax protein in hippocampal neurons. When the ratio of Bcl-2 and Bcl-2 decreased significantly, the apoptosis of hippocampal neurons could be promoted. These results indicate that apoptosis-controlling genes Bcl-2 and Bax in hippocampal neurons play an important role in regulating the apoptosis of cells at different time after exhaustive exercise.
