LYS-84A型离心液相色谱仪(以下称LYS仪),是北京新技术应用研究所研制的一种新型色谱仪。该仪通过装填湿载体物质,利用简单的自动洗脱器进样,在一个特殊的分离盘内,通过离心洗脱从而使混合物得到分离。我们用该仪对二萜醌类、黄酮类物质进行了分离探索,取得较好的结果。一、用LYS仪分离黄酮类物质沙棘总黄酮,主要成份为异鼠李素,槲皮素,异鼠李素-3-鼠李糖甙等七种成份。薄层条件薄层板硅胶G加CMC-N~a(0.1%) 展开剂苯:乙酸乙酯:甲酸(4∶2∶1) (?)板称取35g聚酰胺粉(100-160目),加50%乙醇词成稀糊状铺板(选用直径3mm的垫圈)。上样取300mg总黄酮,溶于无水醇溶液中,待上述板(?)好后,即可用注射器进样。分离进样后,分别用50%乙醇,75%乙醇,95%乙醇溶液洗脱,收集洗脱液30份。
LYS-84A centrifugal liquid chromatography (hereinafter referred to as LYS instrument) is a new type of chromatograph developed by Beijing New Technology Application Research Institute. The device is loaded with a wet carrier material and injected using a simple automatic elution device. The mixture is separated by centrifugation in a special separation tray. We used this instrument to separate and explore diterpenoids and flavonoids and achieved good results. First, the use of LYS apparatus to isolate flavonoids Hippophae rhamnoides total flavonoids, the main ingredients of isorhamnetin, quercetin, isorhamnetin-3-rhamnoside and other seven components. Thin layer condition Silica gel G with CMC-N~a (0.1%) Developing agent Benzene: Ethyl acetate: Formic acid (4:2:1) (?) Plate Weigh 35g of polyamide powder (100-160 mesh) , Add 50% ethanol word into a thin paste board (use a washer with a diameter of 3mm). Take 300mg of total flavonoids, dissolved in anhydrous alcohol solution, after the above plate (?) Well, you can use a syringe injection. After separation and injection, they were eluted with 50% ethanol, 75% ethanol and 95% ethanol respectively, and 30 fractions of the eluate were collected.
