目的对1名AB型献血者血液中的抗-Mur抗体系统地进行特异性、反应能力等检测,并利用此抗体对中山市人群Mur抗原发生频率进行调查。方法在本实验室,应用合成的Mur抗原特异性多肽,通过红细胞多肽凝集抑制试验鉴定抗-Mur;英国国际血型参比实验室对该抗体进行了经典血型血清学试验、试管凝集方法和Miltenberger抗原谱细胞鉴定抗-Mur。观察蛋白水解酶、酸碱度对Mur抗原抗体反应的影响,以微量板盐水法检测广东中山地区Mur抗原发生频率。结果确定了该抗体是高质量的Mur血型抗原检测试剂;木瓜酶、菠萝酶能破坏Mur抗原,弱酸性环境能增强Mur抗原抗体反应,中山地区献血者Mur抗原占7.31%(128/1 752)。结论我们发现的这例人源抗-Mur,无ABO天然抗体存在,效价高,特异性强,为理想的Mur血型定型抗体,为今后国内外输血界对Mur血型相关研究提供了基本保证。中山地区Mur抗原发生频率较高,输血时应该予以充分的重视。
Objective To test the specificity and responsiveness of anti-Mur antibody in the blood of one type AB blood donor and to investigate the frequency of Mur antigen in Zhongshan population using this antibody. Methods In this laboratory, the anti-Mur antibody was synthesized by the Mur antigen-specific peptide. The blood samples were collected from the international blood group reference laboratories of UK with classical serological test, tube agglutination test and Miltenberger antigen Spectral cell identification anti-Mur. To observe the effect of protease and pH on the Mur antigen reaction, and detect the frequency of Mur antigen in Zhongshan district of Guangdong by microplate saline method. As a result, it was confirmed that the antibody was a high quality Mur blood type antigen detection reagent; Papaya enzyme and pineapple enzyme could destroy Mur antigen, and weak acid environment could enhance Mur antigen / antibody reaction. The Mur antigen of Zhongshan blood donors accounted for 7.31% (128/1 752) . Conclusion We found that this human anti-Mur, ABO natural antibodies in the presence of high titer and specificity of the ideal Mur blood typing antibodies for the blood transfusion community at home and abroad on Mur blood related studies provide a basic guarantee. Mur antigen in Zhongshan higher frequency, blood transfusion should be given full attention.
