从中国沙棘茎中提取5-羟色胺(5-HT)是替代动物原料中提取5-HT的最佳途径之一。应用10种大孔吸附树脂对中国沙棘茎5-HT吸附性能筛选,确定HPD树脂对沙棘5-HT具有良好的吸附分离性能。其纯化沙棘5-HT的适宜工艺条件为:乙醇提取物的上样浓度为2.8μg/mL,原液(pH3.9)上样,树脂柱径高比1:5,上样量与树脂体积比1:5。以4倍树脂体积的蒸馏水清洗树脂柱后,用10%乙醇洗脱,洗脱剂用量为4倍量树脂体积,洗脱流速4 BV/h。使用95%乙醇和2%盐酸纯化得到的5-HT的得率为54.67%。
Extraction of 5-hydroxytryptamine (5-HT) from Chinese seabuckthorn stems is one of the best ways to replace 5-HT in animal feedstocks. Ten macroporous adsorption resins were used to screen the 5-HT of Hippophae rhamnoides L. The results showed that HPD resin had good adsorption and separation ability to 5-HT. The optimum conditions for the purification of Hippophae rhamnoides 5-HT were as follows: the sample concentration of ethanol extract was 2.8 μg / mL, the original solution (pH 3.9) was loaded, the resin column height ratio was 1: 5, 1: 5. After the resin column was washed with 4 volumes of distilled water, the column was eluted with 10% ethanol. The amount of eluent used was 4 times the volume of the resin and the elution flow rate was 4 BV / h. The yield of 5-HT obtained using 95% ethanol and 2% hydrochloric acid was 54.67%.
