目的:研究中国苏皖地区汉族人群IL-6-572C>G多态性与前列腺癌易感性的关系。方法:采用病例-对照研究,提取200例前列腺癌患者(病例组)和279例健康体检的非肿瘤患者(对照组)外周血基因组DNA,应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)分析两组IL-6-572C>G位点的多态性,比较不同基因型与前列腺癌易感性的关系。结果:以携带CC基因型的对象为参照组,携带GG基因型者患前列腺癌的风险提高146%(OR=2.46,95%C I:1.41~4.29)。而携带GG基因型者患前列腺癌的风险是携带GC/CC基因型的2.47倍(95%C I:1.47~4.17)。在高龄(>70岁)、高体重指数(>23 kg/m2)、非吸烟、饮酒和有肿瘤家族史5组人群中,携带GG基因型的个体发病风险显著升高,调整后的OR(95%C I)分别为3.06(1.44~6.49)、3.72(1.79~7.74)、2.96(1.30~6.72)、2.73(1.28~5.79)、6.67(1.50~29.69)。结论:IL-6-572C>G与苏皖地区汉族人群前列腺癌的易感性有关,GG基因型可能是前列腺癌的易感基因型。
Objective: To study the relationship between polymorphism of IL-6-572C> G and susceptibility to prostate cancer in Han population of Jiangsu and Anhui provinces. Methods: Genomic DNA was extracted from 200 patients with prostate cancer (case group) and 279 healthy controls (control group) using the case-control study. The genomic DNA was extracted from the peripheral blood by polymerase chain reaction-restriction fragment length polymorphism PCR-RFLP) was used to analyze the polymorphism of IL-6-572C> G locus in two groups and the relationship between different genotypes and susceptibility to prostate cancer was compared. Results: The risk of prostate cancer with GG genotype increased by 146% (OR = 2.46, 95% CI: 1.41-4.29) with reference to subjects with CC genotype. Patients with GG genotype had 2.47 times more risk of prostate cancer (95% CI: 1.47-4.17) than genotype GC / CC. In the elderly (> 70 years old), high body mass index (> 23 kg / m2), non-smoking, alcohol consumption and family history of cancer, individuals with GG genotype had a significantly higher risk of developing disease. 95% CI) were 3.06 (1.44 ~ 6.49), 3.72 (1.79 ~ 7.74), 2.96 (1.30 ~ 6.72), 2.73 (1.28 ~ 5.79) and 6.67 (1.50 ~ 29.69), respectively. Conclusion: IL-6-572C> G is associated with the susceptibility to prostate cancer in Han population in Jiangsu and Anhui Provinces. GG genotype may be the predisposing gene for prostate cancer.
