目的:探讨溃疡性结肠炎虚实标本证候(湿热内蕴证、脾胃虚弱证、脾虚湿热证)之间的microRNA差异表达谱.方法:运用基因芯片技术对采集的溃疡性结肠炎湿热内蕴证(5例)、脾胃虚弱证(5例)、脾虚湿热证(5例)患者及健康志愿者(正常组,5名)的血液标本进行microRNA检测,运用SAS系统对microRNA进行筛选及功能分析.结果:脾胃虚弱证组和正常组筛选出差异表达的microRNA共有45条,29条上调,16条下调;湿热内蕴证组和正常组筛选出差异表达的microRNA共30条,19条上调,11条下调;脾虚湿热证组和正常组筛选出差异表达的microRNA共47条,31条上调,16条下调.脾胃虚弱证组和湿热内蕴证组共筛选出6条差异表达:miR-199a-5p、miR-151-5p、miR-126、miR-532-3p、miR-340、miR-505,差异有统计学意义(P＜0.05),其中上调5条,下调1条;脾胃虚弱证组和脾虚湿热证纽共筛选出5条差异表达:miR-155、miR-21、miR-28-5p、miR-362-3p、miR-199a-3p,差异有统计学意义(P＜0.05),其中上调4条,下调1条;湿热内蕴证组和脾虚湿热证组共筛选出5条差异表达:miR-340、miR-532-3p、miR-318-3p、miR-25、miR-320a,差异有统计学意义(P＜0.05),其中上调4条,下调1条.结论:溃疡性结肠炎虚实标本证候(湿热内蕴证、脾胃虚弱证、脾虚湿热证)患者血浆中存在差异表达microRNA(谱).“,”Objective:To investigate the difference between the expression of microRNA spectrum in ulcerative colitis patients,and the correlation with Biaoben-Xushi symptoms (dampness heat syndrome,spleen stomach deficiency syndrome,spleen deficiency and dampness heat syndrome).Methods:Gene chip technology was used to exam the microRNA expression in patients of ulcerative colitis with dampness heat syndrome (5 cases),spleen and stomach deficiency syndrome (5 cases),and spleen deficiency and dampness heat syndrome (5 cases),and healthy volunteers (normal group,5) were arranged as control.SAS system was used to screen and analyze the microRNA function.Results:The 45 microRNAs was screened out and differentially expressed in spleen and stomach deficiency syndrome and normal group,which was 29 up and 16 down.In dampness heat syndrome and normal group,30 microRNAs was screened out,and 19 up and 11 down.In spleen deficiency and dampness heat syndrome and normal group,47 microRNAs was screened out,and 31 up and 16 down.The 6 differentially expressed miR-199a-5p,miR-151-5p,miR-126,miR-532-3p,miR-340,miR-505 were screened out in dampness heat syndrome and spleen and stomach deficiency syndrome group,and the difference was statistically significant (P＜0.05),of which 5 were up-regulated and 1 down-regulated.Spleen and stomach deficiency group and spleen deficiency syndrome dampness heat group were screened 5 differentially expressed miR-155,miR-21,miR-28-5p,miR-362-3p,miR-199a-3p,and the difference was statistically significant (P＜0.05),of which 4 were up-regulated and 1 down-regulated.The damp heat syndrome group and spleen deficiency dampness heat syndrome group were screened 5 differentially expressed miR-340,miR-532-3p,miR-318-3p,miR-25,miR-320a,and the difference was statistically significant (P＜0.05),of which 4 were up-regulated and 1 down-regulated.Conclusion:MicroRNA differential expression profiles exist in the Biaoben-Xushi syndrome (dampness heat syndrome,spleen stomach deficiency syndrome,spleen deficiency and dampness heat syndrome) of Ulcerative colitis.
