目的:观察丹参注射液联合重组人粒细胞集落刺激因子(rhG-CSF)对急性脑梗死大鼠双侧海马齿状回(DG)干细胞(stem cells)增殖的影响。方法:采用大脑中动脉线栓法制作左侧大脑中动脉闭塞(MCAO)脑梗死大鼠模型,并随机分为丹参注射液组、rhG-CSF组、rhG-CSF+丹参注射液组、生理盐水组,每组6只。按实验方案给药后,每组在干预前及干预7 d后,采用改良神经功能损害评分(m NSS)综合评价神经系统损伤的严重程度;另随机选择6只正常大鼠作为假手术组。以免疫荧光染色法检测DG区与增殖细胞相关的Ki67(Ki67)/巢蛋白(nestin)、造血干细胞(CD34+)阳性细胞数。结果:rhG-CSF+丹参注射液组大鼠m NSS评分低于丹参注射液组、rhG-CSF组、生理盐水组,差异均有统计学意义(P<0.05);rhG-CSF+丹参注射液组DG的Ki67+/nestin+细胞数高于其余4组。rhG-CSF组及rhG-CSF+丹参注射液组在脑梗死侧均发现CD34+。结论:丹参注射液联合rhG-CSF能促进脑梗死大鼠海马齿状回神经新生,改善脑梗死大鼠的神经功能,可能在脑梗死后神经功能恢复过程中发挥了重要作用。
OBJECTIVE: To observe the effect of Salvia miltiorrhiza injection combined with recombinant human granulocyte colony-stimulating factor (rhG-CSF) on the proliferation of stem cells of bilateral hippocampal dentate gyrus (DG) in rats with acute cerebral infarction. METHODS: The middle cerebral artery occlusion (MCAO) cerebral infarction model was established by middle cerebral artery occlusion and randomly divided into Danshen injection group, rhG-CSF group, rhG-CSF+ Danshen injection group, and saline group. , 6 per group. After administration according to the experimental protocol, the severity of nervous system injury was comprehensively evaluated by Modified Neural Impairment Score (m NSS) before intervention and 7 days after intervention. Another 6 normal rats were randomly selected as sham operation group. The number of Ki67 (ki67)/nestin and hematopoietic stem cells (CD34+) positive cells associated with proliferating cells in the DG area was detected by immunofluorescence staining. Results: The scores of m NSS in rhG-CSF+Danshen injection group were lower than those in Danshen injection group, rhG-CSF group, and saline group (P<0.05); rhG-CSF+Danshen Injection group DG The number of Ki67+/nestin+ cells was higher than the remaining 4 groups. RhG-CSF group and rhG-CSF + Salvia miltiorrhiza injection group were found in the cerebral infarction CD34 +. CONCLUSION: Salvia miltiorrhiza injection combined with rhG-CSF can promote neurodevelopment of the dentate gyrus in hippocampus of rats with cerebral infarction and improve neurological function in cerebral infarction rats. It may play an important role in the recovery of neurological function after cerebral infarction.
