目的探讨膀胱移行细胞癌(BTCC)组织中E-钙黏附蛋白(CDH1)表达及其与BTCC发病机制间的相关性。方法 BTCC患者36例,分别取血液、肿瘤、癌旁组织标本。对照组30例,取血液标本。ELISA法测定血浆中CDH1,免疫组化法检测组织CDH1的表达。竞争性PCR方法半定量分析组织中CDH1表达。结果 BTCC组血浆中CDH1(3.4±1.6)ng/ml较非肿瘤组(6.3±2.7)ng/ml低(P<0.05)。免疫组化示BTCC组织中CDH1表达阳性率为33.3%(11/36),癌旁组织中的表达阳性率为63.8%(23/36),BTCC组织CDH1mRNA水平与癌旁组织比较差异有统计学意义(P<0.05)。结论 CDH1基因表达下调可能在BTCC发生中发挥重要作用。
Objective To investigate the expression of E-cadherin (CDH1) in bladder transitional cell carcinoma (BTCC) and its relationship with the pathogenesis of BTCC. Methods Thirty-six patients with BTCC were divided into three groups: blood, tumor and paracancerous tissues. Control group of 30 cases, take blood samples. The level of CDH1 in plasma was measured by ELISA and the expression of CDH1 was detected by immunohistochemistry. Competitive PCR method Semi-quantitative analysis of CDH1 expression in tissues. Results The plasma levels of CDH1 (3.4 ± 1.6) ng / ml in BTCC group were significantly lower than those in non-tumor group (6.3 ± 2.7) ng / ml group (P <0.05). Immunohistochemistry showed that the positive rate of CDH1 expression in BTCC tissues was 33.3% (11/36), the positive expression rate in cancerous tissues was 63.8% (23/36). The difference between CDC1 mRNA expression in BTCC tissues and adjacent non-cancerous tissues was statistically significant Significance (P <0.05). Conclusion The down-regulation of CDH1 gene may play an important role in the occurrence of BTCC.
