目的用卵白蛋白(OVA)诱导建立的急性支气管哮喘大鼠模型,研究神经生长因子(NGF)处理后肺部粘蛋白基因Muc5ac的表达,探讨神经生长因子在哮喘发病中的作用。方法 27只SD大鼠随机分为正常对照组、哮喘组和NGF干预组。激发后24 h解剖大鼠,取肺组织用免疫组化染色法测定大鼠肺中Muc5ac表达情况。结果正常对照组、哮喘组及NGF干预组肺Muc5ac蛋白含量(用阳性面积μm2表示)分别为(501.8±270.6)、(1 330.2±662.9)和(2 033.9±892.3)。结论哮喘组大鼠粘蛋白基因Muc5ac表达增加,NGF加重哮喘气道炎症,在哮喘发病中起一定作用。
OBJECTIVE: To establish a rat model of acute bronchial asthma induced by ovalbumin (OVA) and to study the expression of Muc5ac in lung mucosa after NGF treatment and to explore the role of NGF in the pathogenesis of asthma. Methods Twenty-seven SD rats were randomly divided into normal control group, asthma group and NGF intervention group. Rats were dissected 24 h after challenge and Muc5ac expression in lungs of rats was determined by immunohistochemical staining. Results The levels of Muc5ac protein in the normal control group, asthma group and NGF intervention group were (501.8 ± 270.6), (1 330.2 ± 662.9) and (2 033.9 ± 892.3), respectively. Conclusions The Muc5ac expression of mucin gene is increased in asthmatic rats. NGF aggravates airway inflammation in asthmatic rats and plays a role in the pathogenesis of asthma.
