锡金海棠[Malus sikkimensis(Wenz.)Koehne]是我国稀有植物和优良苹果砧木种质资源。本研究探讨了影响锡金海棠离体再生的条件,建立了锡金海棠子叶高效离体再生体系。结果表明,子叶再生体系的最佳诱导培养基为SC+0.1mg·L-1NAA+2.0mg·L-1TDZ,最佳分化培养基为SC+0.1mg·L-1NAA+1.5mg·L-1TDZ,最佳继代增殖培养基为SC+2.0mg·L-16-BA+0.2mg·L-1NAA,最佳生根培养基为1/2MS+1.0mg·L-1IBA。炼苗后,移入珍珠岩、花生壳与草炭(1:1:1)的基质中,20d移栽成活率高达90%。
Malus sikkimensis (Wenz.) Koehne is a rare germplasm resource and an apple rootstock germplasm in China. In this study, we studied the conditions that affect the regeneration of Siniperca chuatsi and established an efficient regeneration system in vitro. The results showed that the optimum culture medium for cotyledon regeneration system was SC + 0.1 mg · L-1NAA + 2.0 mg · L-1TDZ, and the best differentiation medium was SC + 0.1 mg · L-1 NAA + 1.5 mg · L-1TDZ , The best subculture medium was SC + 2.0mg · L-16-BA + 0.2mg · L-1NAA, the best rooting medium was 1 / 2MS + 1.0mg · L-1IBA. After refining, into the perlite, peanut shells and peat (1: 1: 1) matrix, 20d transplanting survival rate of up to 90%.
