目的研究激活诱导细胞死亡(AICD)对尖锐湿疣患者细胞免疫功能的影响。方法分离CA 患者和健康体检正常人的外周血单个核细胞(PBMC),在PHA的刺激下体外培养48h后收集细胞和上清液,经流式细胞术检测凋亡细胞,应用酶联免吸附试验检测培养上清液IL-2 及IL-10。结果CA组新鲜血PBMC的凋亡率很低和对照组相似(P>0.05),培养48h后CA 患者的外周血单个核细胞凋亡明显高于正常对照组(P<0.001),CA患者的培养上清液IL-2 浓度明显低于对照组(P<0.001),而IL-10浓度显著高于对照组(P<0.001)。结论AICD可能是导致尖锐湿疣患者细胞免疫功能低下的一个重要原因,在尖锐湿疣患者发病机理中起重要作用。
Objective To investigate the effect of AICD on cellular immune function in patients with condyloma acuminatum. Methods Peripheral blood mononuclear cells (PBMCs) from patients with CA and healthy controls were isolated and cultured in vitro for 48 h after PHA stimulation. Cells were harvested and supernatants were harvested. Apoptotic cells were detected by flow cytometry. Test culture supernatant IL-2 and IL-10. Results The apoptosis rate of PBMC in fresh blood was very low in CA group (P> 0.05). After 48 hours of culture, the apoptosis of peripheral blood mononuclear cells in CA group was significantly higher than that in normal control group (P <0.001) The concentration of IL-2 in culture supernatant was significantly lower than that in control group (P <0.001), while the concentration of IL-10 in supernatant was significantly higher than that in control group (P <0.001). Conclusion AICD may be one of the important causes of cellular immune dysfunction in patients with condyloma acuminatum and play an important role in the pathogenesis of condyloma acuminatum.
