目的:采用二次回归通用旋转试验设计优化土茯苓总黄酮(TFSG)的提取工艺。方法:根据Box-Be-hnken的中心组合试验设计原理,在单因素试验的基础上采用四因素三水平的响应曲面分析法,依据回归分析确定各工艺条件的影响因子,以TFSG得率(%)为响应值作响应面和等高线图。结果:TFSG超声波辅助提取效果的影响因素由大到小顺序为:乙醇浓度>超声功率>超声时间>料液比。优化后的工艺参数为:乙醇浓度57%,超声功率500W,超声时间34min,料液比1:21,响应值达到2.32%。结论:本文所建立的数学模型极其显著(P<0.01),可用来对TFSG的提取工艺进行分析和预测。
Objective: To optimize the extraction process of TFSG by using quadratic regression general rotation test. Methods: According to the design principle of Box-Be-Hnken’s central composite experiment, four factors and three levels of response surface analysis were used on the basis of single-factor experiment, and the influencing factors of each technological condition were determined by regression analysis. The TFSG yield Respond to the response surface and contour plots. Results: The order of influencing factors of ultrasonic assisted extraction of TFSG was as follows: ethanol concentration> ultrasonic power> ultrasonic time> solid-liquid ratio. The optimized process parameters are: ethanol concentration 57%, ultrasonic power 500W, ultrasonic time 34min, material-liquid ratio 1:21 and response value 2.32%. Conclusion: The mathematical model established in this paper is extremely significant (P <0.01), which can be used to analyze and predict TFSG extraction process.
