目的:观察SD大鼠胰岛β细胞G蛋白偶联受体40(GPR40)的表达与内脏脂肪含量及胰岛素1相分泌之间的关系。方法:大鼠按体质量分为3组(100g、200g及300g组),行静脉糖耐量实验,胶原酶原位灌注法分离胰岛,免疫组织化学法结合激光扫描共聚焦显微镜技术对胰岛β细胞表达的GPR40进行定位,并行半定量分析。结果:随着大鼠体质量的增加,内脏脂肪含量明显增加,300g组大鼠胰岛β细胞GPR40表达较100g及200g组明显增加,3组大鼠胰岛素1相分泌无显著差异。结论:GPR40表达的变化可能与内脏脂肪含量及年龄因素有关,其表达水平对正常大鼠葡萄糖刺激的胰岛素分泌无明显影响。
OBJECTIVE: To observe the relationship between the expression of G protein-coupled receptor 40 (GPR40) and visceral fat content and insulin 1 phase secretion in SD rats. Methods: The rats were divided into 3 groups (100g, 200g and 300g groups) according to their body weight. Intravenous glucose tolerance test and collagenase in situ perfusion were used to isolate islets. Immunohistochemistry and laser scanning confocal microscope The expressed GPR40 was localized and subjected to semi-quantitative analysis in parallel. Results: With the increase of body weight, the content of visceral fat increased obviously. The expression of GPR40 of islet β cells of 300g group increased obviously compared with that of 100g and 200g group. There was no significant difference of insulin secretion among 3 groups. CONCLUSION: The changes of GPR40 expression may be related to visceral fat content and age, and its expression level has no significant effect on glucose-stimulated insulin secretion in normal rats.
