目的:探讨中国人低密度脂蛋白受体(LDL—R)基因NcoI多态性及其与血脂含量的关系。方法:合成引物一对,用PCR技术扩增LDL—R基因外显子18的特异片段,用限制性内切酶NcoI酶切,得N+N+、N+N-、N-N-3种基因型。结果:沈阳地区102例健康汉族人的LDL—R基因NcoI多态性N+N+占44.1%、N+N-占45.1%、N-N-占10.8%。N+等位基因频率0.667,N-等位基因频率0.333。N+N+、N+N-、N-N-3组的血清总胆固醇含量依次为4.78mmol/L、4.16mmol/L、3.65mmol/L,差异显著(F=8.17P<0.05)。结论:提示N+等位基因可能与高胆固醇有关系
Objective: To investigate the NcoI polymorphism of Chinese low density lipoprotein receptor (LDL-R) gene and its relationship with blood lipid content. Methods: A pair of primers was synthesized. The specific fragment of exon 18 of LDL-R gene was amplified by PCR and was digested with restriction endonuclease NcoI to get N + N +, N + N- and N-N-3 genotypes. Results: The NcoI polymorphism of LDL-R gene in 102 healthy Han nationality in Shenyang was 44.1%. N + N accounted for 45.1% and N-N accounted for 10.8%. The N + allele frequency was 0.667 and the N-allele frequency was 0.333. The levels of total cholesterol in N + N +, N + N- and N-N-3 groups were 4.78mmol / L, 4.16mmol / L and 3.65mmol / L respectively, with significant difference (F = 8.17P <0.05). Conclusion: It suggests that the N + allele may be related to high cholesterol
