目的:优选猪苓液体发酵生产胞内多糖的工艺条件。方法:采用摇瓶培养法制备猪苓胞内多糖的发酵培养基,以猪苓胞内多糖得率为指标,通过单因素试验考察氮源、碳源和发酵条件,正交试验优选培养基的组成。结果:液体发酵生产猪苓胞内多糖的最适碳源为蔗糖,最适氮源为黄豆粉,优选的培养基组成为马铃薯10.0%,蔗糖4.0%,黄豆粉0.3%,KH2PO40.2%,MgSO4·7H2O 0.15%;发酵条件为初始pH 5.2~5.6,振荡速度180~200 r·min-1,培养温度28℃,培养时间7 d。结论:液体发酵猪苓胞内多糖具有周期短、产量高的优势,对猪苓多糖工业化生产和降低生产成本具有重要意义。
Objective: To optimize the fermentation conditions of Polysaccharide from Polyporus umbellatus by liquid fermentation. Methods: The fermentation medium of Polyporus umbellatus polysaccharides was prepared by shake flask culture. The yield of Polyporus umbellatus polysaccharides was used as an index. The nitrogen source, carbon source and fermentation conditions were investigated by single factor experiment. composition. Results: The most suitable carbon source for the production of Polyporus umbellatus by liquid fermentation was sucrose. The optimum nitrogen source was soybean flour. The optimal medium consisted of 10.0% of potato, 4.0% of sucrose, 0.3% of soybean flour, 40.2% of KH2PO, MgSO4 · 7H2O 0.15%. The fermentation conditions were initial pH 5.2 ~ 5.6, shaking speed 180 ~ 200 r · min-1, culture temperature 28 ℃ and culture time 7 d. Conclusion: Liquid fermentation of Polyporus umbellatus polysaccharides has the advantages of short cycle and high yield, which is of great significance for the industrial production of Polyporus umbellatus polysaccharide and the reduction of production cost.
