采用光合菌适用的富集培养基,通过富集驯化的培养方法,从养猪场废水污泥中采集菌液进行较长时间的驯化富集,分离纯化出A2光合菌株,为红色单菌落,革兰氏染色为阴性。对A2菌株生长特性研究表明,添加1.0%Na HCO3能使A2菌株能较早进入生长对数期和稳定期。A2菌株的最适培养条件为:p H为7.5、30℃厌氧光照培养。将菌株A2处理模拟污水和猪场实际污水,探讨菌株A2对氮、磷的去除效果。结果表明,A2菌株处理模拟污水8 d,对TN、NH+4-N、NO-3-N、NO-2-N和TP的平均去除率分别为:74.38%、91.09%、56.17%、90.76%和57.82%。A2菌株处理猪场污水8d,对TN、NH+4-N、NO-3-N、NO-2-N和TP的平均去除率分别为:52.45%、31.19%、45.30%、88.63%和50.92%。表明,菌株A2具有良好的污水处理应用价值。
Using the enrichment culture medium suitable for photosynthetic bacteria, the bacterial liquid was collected from piggery wastewater sludge for domestication and enrichment for a long time through enrichment and acclimation culture method. The A2 photosynthetic bacteria were isolated and purified as single red colonies, Gram stain is negative. The growth characteristics of A2 strain showed that the addition of 1.0% Na HCO3 can strain A2 earlier into the logarithmic growth phase and stationary phase. The optimal culture conditions for A2 strain were as follows: p H 7.5, anaerobic light culture at 30 ℃. The strain A2 was used to treat the simulated sewage and the actual sewage in pig farms to explore the effect of strain A2 on nitrogen and phosphorus removal. The results showed that the average removal rates of TN, NH + 4-N, NO-3-N, NO-2-N and TP by A2 strain were 74.38%, 91.09%, 56.17% and 90.76 % And 57.82%. The average removal rates of TN, NH + 4-N, NO-3-N, NO-2-N and TP by A2 strain were respectively 52.45%, 31.19%, 45.30%, 88.63% and 50.92 %. Show that strain A2 has a good sewage treatment value.
