目的探讨脐带间充质干细胞(UC-MSCs)的甲基纤维素涂剂对兔角膜损伤的修复作用。方法体外进行UC-MSCs的培养和鉴定,采用NaOH溶液制作兔角膜烧伤模型,烧伤后以甲基纤维素为载体将UC-MSCs移植到烧伤的兔角膜表面,以单纯甲基纤维素组及碱烧伤组作为对照组,在裂隙灯下观察角膜的临床改变。结果成功地培养出UC-MSCs;碱烧伤阳性对照组和单纯甲基纤维素组均可见角膜浑浊呈瓷白色、表面粗糙,不能透见眼内结构。UC-MSCs甲基纤维素组,新生血管较少,角膜表面较光滑,角膜透明度明显改善。结论 UC-MSCs可以在兔角膜表面成活并向角膜上皮细胞分化,UC-MSCs的甲基纤维素凃剂对碱烧伤的角膜损伤有一定的修复作用。
Objective To investigate the effect of methylcellulose coated umbilical cord mesenchymal stem cells (UC-MSCs) on corneal injury in rabbits. Methods UC-MSCs were cultured and identified in vitro. Rabbit corneal burn model was made by NaOH solution. After burn, UC-MSCs were transplanted onto the surface of burned rabbit cornea with methylcellulose as carrier. Burns group as a control group, in the slit lamp to observe the clinical changes of the cornea. Results UC-MSCs were successfully cultured. Alkali burn positive control group and methylcellulose alone group showed corneal opacity was porcelain white with rough surface, which could not be seen through intraocular structures. UC-MSCs methylcellulose group, less angiogenesis, corneal surface is smooth, corneal transparency was significantly improved. Conclusion UC-MSCs can survive on corneal surface of rabbit and differentiate into corneal epithelial cells. Methylcellulose of UC-MSCs can repair corneal injury of alkali burns.
