目的 采用网络药理学方法研究黄连-三七药对抗2型糖尿病的作用机制.方法 通过检索中药系统药理数据分析平台(TCMSP),获得黄连、三七主要活性成分和相关作用靶点,并采用Cytoscape 3.6.1版软件构建黄连-三七药对活性成分-靶点网络图.通过比较毒物基因组学数据库(CTD)和靶向治疗数据库(TTD)中检索抗2型糖尿病靶点,在STRING数据库进行蛋白质间相互作用分析,构建抗2型糖尿病把点PPI网络图,利用Cytoscape 3.6.1版软件中Merge功能与活性成分-靶点融合,获得黄连-三七药对抗2型糖尿病作用靶点.运用注释、可视化和集成发现数据库(DAVID)平台对靶点进行基因本体(GO)功能富集分析和京都基因与基因组百科全书(KEGG)信号通路富集分析,进一步研究其作用机制.结果 获得具有类药性、口服吸收良好的成分17个,黄连-三七药对抗2型糖尿病作用靶点19个,包括肠道麦芽糖酶-葡糖淀粉酶(MGAM)、二肽基肽酶Ⅳ(DPP4)、胰岛素受体(INSR)、过氧化物酶体增殖物激活受体γ(PPARG)等.GO分析得到GO条目111个(P<0.05),其中生物过程(BP)条目92个,细胞组成(CC)条目12个,分子功能(MF)条目7个.KEGG分析得到信号通路21条(P<0.05),包括HIF-1信号通路、胰岛素抵抗通路、脂肪细胞脂解调控通路、PI3K-Akt信号通路等.结论 黄连-三七的活性成分主要通过多把点、多通路起到抗2型糖尿病的作用,为深入解析黄连-三七药对复杂体系,阐释其抗2型糖尿病的潜在复杂作用机制提供了新方向.
