凋亡是帕金森病多巴胺能神经元损伤的形式之一。本文概述了前凋亡分子CHOP/GADD153的结构、分布、功能及表达调控,它是内质网应激相关凋亡通路上的重要分子。在帕金森病发病时其表达上调,是细胞由防御性应激反应向凋亡转变的促发因素。另外,CHOP/GADD153还参与调节多个与凋亡相关分子和通路,故其作为潜在的帕金森病神经治疗靶点之一,对深入研究帕金森病细胞凋亡早期的分子机制和内质网应激与帕金森病发病的联系提供了新的思路。
Apoptosis is one of the forms of dopaminergic neuron damage in Parkinson’s disease. This review summarizes the structure, distribution, function and expression of the pro-apoptotic molecule CHOP / GADD153, which is an important molecule in the ER stress-related apoptotic pathway. In the pathogenesis of Parkinson’s disease, its expression is up-regulated, which is the trigger factor of cell transformation from defensive stress response to apoptosis. In addition, CHOP / GADD153 is also involved in the regulation of multiple apoptosis-related molecules and pathways. Therefore, CHOP / GADD153, as one of potential neuropathies for Parkinson’s disease, is useful in further studying the molecular mechanisms and endoplasmic reticulum Stress and the pathogenesis of Parkinson’s disease provides a new way of thinking.
