研究电离辐射在离体时诱发人细胞恶性转化。方法选用α辐射源诱发SV40永生化的人胎气管成纤维细胞恶性转化,观察在细胞恶性转化过程中细胞生物学特性的变化。结果经α粒子照射后细胞形态发生转化;细胞相对存活率与剂量呈依赖性,随着照射剂量的增加,存活率呈指数下降,存活曲线呈单击单靶型;早期传代细胞染色体数出现非整倍体和非稳定性畸变,同时有两个标记染色体;软琼脂克隆形成率随着照后传代次数的增加而明显增高;第40代细胞接种裸鼠长出肿瘤结节,病理诊断为纤维肉瘤。结论α粒子可诱发人胎永生化气管成纤维细胞体外恶性转化。
Study of ionizing radiation in vitro induced malignant transformation of human cells. Methods The malignant transformation of SV40 immortalized human tracheal fibroblasts was induced by α radiation and the changes of cell biological characteristics during malignant transformation of cells were observed. Results The morphological changes of cells were observed after exposure to α particles. The relative survival rate was dose-dependent. The survival rate decreased exponentially with the increase of irradiation dose, and the survival curve was single-target type. The chromosome number of early passaged cells Euploid and non-stable aberrations, with two marker chromosomes. The rate of colony formation of soft agar increased with the increase of the number of passages after irradiation. The 40th generation cells inoculated with nude mice developed tumor nodules and the pathological diagnosis was fiber sarcoma. Conclusion α particles can induce malignant transformation of human fetal immortalized tracheal fibroblasts in vitro.
