目的探讨限食(CR)和n-6、n-3多不饱和脂肪酸(PUFA)对大鼠溃疡性结肠炎模型的影响,以制备溃疡性结肠炎。方法 40只SD雌性大鼠按体重随机分为5组,即对照组、模型组、限食组、5∶1 PUFA自由进食组、5∶1 PUFA限食组。限食组喂食量是自由进食组的60%。喂养14周后,除对照组外,其余组给予5%葡聚糖硫酸钠(DSS)水溶液,自由饮用7 d造模。结果与自由进食组相比,限食组体重增长缓慢,尤其是5∶1PUFA限食组大鼠体重最轻(P<0.05)。与模型组比,5∶1 PUFA限食组大鼠的血清MPO活性、疾病活动指数、结肠大体和组织病理学评分降低(P<0.05)。与限食组比较,5∶1 PUFA限食组大鼠血清IL-6和TNF-α含量均降低,而IL-10含量升高(P<0.05)。结论膳食中添加n-6与n-3比例为5∶1的PUFA和限食联合作用对DSS诱导的大鼠结肠炎有明显保护作用。
Objective To investigate the effects of food restriction (CR) and n-6, n-3 polyunsaturated fatty acids (PUFA) on the ulcerative colitis model in rats to prepare ulcerative colitis. Methods 40 SD female rats were randomly divided into 5 groups according to body weight: control group, model group, restricted diet group, 5: 1 PUFA free diet group and 5: 1 PUFA restricted diet group. Food restriction group feeding is 60% of the group fed freely. After 14 weeks of feeding, except for the control group, the remaining groups were given 5% dextran sodium sulfate (DSS) aqueous solution and were allowed to drink for 7 days to make model. Results Compared with the free-eating group, the body weight of the limited-feeding group increased slowly, especially in the 5: 1 PUFA fastidious group (P <0.05). Compared with the model group, the serum MPO activity, disease activity index, gross colon and histopathological score of 5: 1 PUFAs group decreased (P <0.05). Compared with the limited diet group, the levels of IL-6 and TNF-α in serum of 5: 1 PUFA group decreased and IL-10 content increased (P <0.05). Conclusion Dietary supplementation of 5: 1 n-6 and n-3 PUFAs and combination of limited diet had significant protective effect on DSS-induced colitis in rats.
