了解血镁浓度与环孢素A(CsA)肾损害之间的关系。方法采用大鼠离体肾灌流模型,观察了Mg2+对CsA的急性肾脏效应的影响。结果正镁(1.2mmol/L)时CsA50mg/L灌流引起显著的肾血管痉挛和肾灌注不良,以及Ca2+聚积和Mg2+耗竭;在低镁(0.12mmol/L)时,CsA的作用明显增强;而高镁(4.8mmol/L)时,CsA的作用受到了有效的抑制。结论适当提高Mg2+浓度能改善离体肾血流动力学和克服CsA对肾脏的影响。鉴于使用CsA的患者常有低镁血症,因此适当补镁对防治CsA肾毒性具有一定意义。
To understand the relationship between serum magnesium concentration and cyclosporin A (CsA) renal damage. Methods The isolated rat renal perfusion model was used to observe the effect of Mg2 + on the acute kidney effect of CsA. RESULTS: CsA 50 mg / L perfusion caused significant renal vasospasm and renal perfusion, as well as accumulation of Ca2 + and depletion of Mg2 + in normal magnesium (1.2 mmol / L). The effect of CsA at low magnesium (0.12 mmol / L) While the effect of CsA was effectively inhibited by high magnesium (4.8 mmol / L). Conclusion Appropriate increase of Mg2 + concentration can improve renal hemodynamics and overcome the influence of CsA on the kidneys. Given that patients with CsA often have hypomagnesemia, proper magnesium supplementation has some significance in preventing and treating CsA nephrotoxicity.
